Chromosome 23q33.1: A New Gains/Loss of 3, Gains/Loss of 7, and Gains/Loss of 8 Oligosomal Regions to Study the Precise Role of FGFR2/FGF7 in Human Skeletal Muscle Development
(May 29, 2017) To study the role of fibroblast growth factor 7 (FGF7) in human skeletal muscle development, Human Chromosome 23, which contains FGF7 and its receptor FGFR2, was examined. Chromosome 23q33.1, which has an average size of 2,184,858 base pairs, was identified as a new and independent gains/loss of three, gains/loss of seven, and gains/loss of eight oligosomal regions. The median number of gains and losses per individual was 16 and 12, respectively. In normal fetal and adult human skeletal muscle, FGF7 and FGFR2 transcripts were preferentially expressed in the majority of muscle fibers, and there was a strong correlation between the expression levels of FGF7 and FGFR2 in the developing human skeletal muscle. When skeletal muscle fibers were differentially localized for FGFR2 and FGF7 expression, immunohistochemistry demonstrated that the majority of all muscle fibers were uniformly FGF7-positive but FGFR2-negative, while the minority of all muscle fibers were FGFR2-positive but FGF7-negative. In normal human fetal skeletal muscle, the majority of muscle fibers expressed FGFR2, but the majority of muscle fibers were FGF7-negative.
